Does Benadryl cause dementia? The famous number is for a whole class

Nobody knows yet. The often-quoted 54% higher dementia rate was in the heaviest users of any strong anticholinergic drug, not antihistamines alone, and the big antihistamine studies disagree.

The Benadryl dementia warning travels with a number: a 54% higher rate of dementia in adults over 65 followed for seven years. That was the heaviest users of every strong anticholinergic drug together, a class that blocks one brain messenger, and antihistamines were under a fifth of it. The two largest studies of antihistamines themselves point opposite ways, and neither can separate the drug from the reason it was taken. The question is open, not settled either way.

Among 284,343 English patients, antihistamines alone showed no significant link at the heaviest exposure. Among 714,052 Taiwanese patients, the sedating ones rose to 51% higher.
